Early brainstem [18F]THK5351 uptake is linked to cortical hyperexcitability in healthy aging

چکیده
BACKGROUND. Neuronal hyperexcitability characterizes the early stages of Alzheimer’s disease (AD). In animals, misfolded tau and amyloid-β (Aβ) protein accumulation — both central to AD neuropathology promote cortical excitability neuronal network dysfunction. healthy humans, Aβ aggregates are first detected, respectively, in brainstem frontomedial temporobasal cortices, decades prior onset cognitive symptoms. Whether is related its associated neuroinflammation aggregations remains unknown.

Interleukin-6 is linked to longitudinal rates of cortical thinning in aging.
Interleukin-6 (IL-6) is a pro-inflammatory cytokine produced by immune cells and other cell types such as microglia throughout the brain. Higher levels of IL-6 in older adults have been cross-sectionally and longitudinally associated with physical and cognitive impairment, as well as increased dementia risk. The association between IL-6 levels and structural and functional brain changes is less...
متن کاملCortical thickness is linked to executive functioning in adulthood and aging.
Executive functions that are dependent upon the frontal-parietal network decline considerably during the course of normal aging. To delineate neuroanatomical correlates of age-related executive impairment, we investigated the relation between cortical thickness and executive functioning in 73 younger (20-32 years) and 56 older (60-71 years) healthy adults. Executive functioning was assessed usi...
متن کاملCognition in healthy aging is related to regional white matter integrity, but not cortical thickness.
It is well established that healthy aging is accompanied by structural changes in many brain regions and functional decline in a number of cognitive domains. The goal of this study was to determine (1) whether the regional distribution of age-related brain changes is similar in gray matter (GM) and white matter (WM) regions, or whether these two tissue types are affected differently by aging, a...
متن کاملCortical hyperexcitability and sensitivity to discomfort glare
It is well established that there are two main aspects to glare, the visual impairment and the discomfort, known as disability and discomfort glare, respectively. In contrast to the case of disability glare we understand very little about the underlying mechanisms or physiology of discomfort glare. This study attempts to elucidate the neural mechanisms involved using fMRI and glare sources with...
متن کاملNovel threshold tracking techniques suggest that cortical hyperexcitability is an early feature of motor neuron disease.
The dying forward hypothesis of motor neuron disease (MND) suggests that corticomotoneurons induce excitotoxic anterior horn cell death, with involvement of the glutamatergic neurotransmitter system. The aim of the present study was to apply novel threshold tracking transcranial magnetic stimulation (TMS) techniques in conjunction with peripheral nerve excitability studies in MND patients to fu...
